The p.S483R variant (also known as c.1449T>G), located in coding exon 11 of the SMAD4 gene, results from a T to G substitution at nucleotide position 1449. The serine at codon 483 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

This sequence change replaces serine, which is neutral and polar, with arginine, which is basic and polar, at codon 483 of the SMAD4 protein (p.Ser483Arg). This variant is present in population databases (rs745598003, gnomAD 0.003%). This variant has not been reported in the literature in individuals affected with SMAD4-related conditions. ClinVar contains an entry for this variant (Variation ID: 529929).
