The p.G2166D variant (also known as c.6497G>A), located in coding exon 12 of the ALPK2 gene, results from a G to A substitution at nucleotide position 6497. The glycine at codon 2166 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
