This variant has not been reported in the literature in individuals with SMARCA4-related conditions. This sequence change replaces proline with alanine at codon 4 of the SMARCA4 protein (p.Pro4Ala). The proline residue is highly conserved and there is a small physicochemical difference between proline and alanine. This variant is not present in population databases (ExAC no frequency). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
Hereditary cancer-predisposing syndrome Uncertain:1
The p.P4A variant (also known as c.10C>G), located in coding exon 1 of the SMARCA4 gene, results from a C to G substitution at nucleotide position 10. The proline at codon 4 is replaced by alanine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this alteration remains unclear. -
