The c.1706-1 G>C pathogenic variant in the LDLR gene has been reported in one child with elevated serum total cholesterol who was part of a national universal screening program for hypercholesterolemia (Klancar et al., 2015). This variant destroys the canonical splice acceptor site in intron 11 and is predicted to cause abnormal gene splicing. This variant is predicted to lead to either an abnormal message that is subject to nonsense-mediated mRNA decay, or to an abnormal protein product if the message is used for protein translation. Other splice site variants in the LDLR gene, including variants in the same nucleotide (c.1706-1 G>T, c.1706-1 G>A), have been reported in HGMD in association with FH (Stenson et al., 2014). Furthermore, the c.1706-1 G>C variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server).
