Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. This variant disrupts the p.Asp601 amino acid residue in LDLR. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 11933210, 20236128, 25461735, 33231818). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. This sequence change replaces aspartic acid, which is acidic and polar, with valine, which is neutral and non-polar, at codon 601 of the LDLR protein (p.Asp601Val). This variant is not present in population databases (gnomAD no frequency).
