This sequence change replaces lysine, which is basic and polar, with arginine, which is basic and polar, at codon 48 of the STK11 protein (p.Lys48Arg). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with STK11-related conditions. ClinVar contains an entry for this variant (Variation ID: 505434). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed at Invitae for this missense variant, however the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on STK11 protein function. Experimental studies have shown that this missense change does not substantially affect STK11 function (PMID: 18687677, 23577667, 30594553, 31217475, 31554794). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
Hereditary cancer-predisposing syndrome Uncertain:2
The p.K48R variant (also known as c.143A>G), located in coding exon 1 of the STK11 gene, results from an A to G substitution at nucleotide position 143. The lysine at codon 48 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
