Variant summary: STK11 c.1193C>T (p.Ala398Val) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 1.3e-05 in 228336 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1193C>T has been reported in the literature as a VUS in settings of multigene panel testing among individuals undergoing testing for hereditary cancer (example, Yurgelun_2015, Caminsky_2016, Bhai_2021), however it was also found in 2/7325 European American women, who were older than 70 years of age, and never had cancer (in the FLOSSIES database). These report(s) do not provide unequivocal conclusions about association of the variant with Peutz-Jeghers Syndrome. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 34326862, 26898890, 25980754). ClinVar contains an entry for this variant (Variation ID: 184646). Based on the evidence outlined above, the variant was classified as uncertain significance. -
