Genes affected
WDR62 (HGNC:24502): (WD repeat domain 62) This gene is proposed to play a role in cerebral cortical development. Mutations in this gene have been associated with microencephaly, cortical malformations, and cognitive disability. Alternative splicing results in multiple transcript variants. This sequence change creates a premature translational stop signal (p.Arg512*) in the WDR62 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in WDR62 are known to be pathogenic (PMID: 20729831). This variant is present in population databases (rs201993064, gnomAD 0.008%). This variant has not been reported in the literature in individuals affected with WDR62-related conditions. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. For these reasons, this variant has been classified as Pathogenic. -
