The p.R41L variant (also known as c.122G>T), located in coding exon 1 of the FKRP gene, results from a G to T substitution at nucleotide position 122. The arginine at codon 41 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear. -

This sequence change replaces arginine, which is basic and polar, with leucine, which is neutral and non-polar, at codon 41 of the FKRP protein (p.Arg41Leu). This variant is present in population databases (rs201497063, gnomAD 0.07%). This variant has not been reported in the literature in individuals affected with FKRP-related conditions. ClinVar contains an entry for this variant (Variation ID: 459228).
