This variant disrupts the p.Arg40 amino acid residue in CRX. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 26161267, 29785639, 31626798, 32533067). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This variant has not been reported in the literature in individuals affected with CRX-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces arginine, which is basic and polar, with proline, which is neutral and non-polar, at codon 40 of the CRX protein (p.Arg40Pro). -
