This sequence change replaces arginine, which is basic and polar, with glycine, which is neutral and non-polar, at codon 180 of the POLD1 protein (p.Arg180Gly). This variant is present in population databases (no rsID available, gnomAD no frequency). This variant has not been reported in the literature in individuals affected with POLD1-related conditions. ClinVar contains an entry for this variant (Variation ID: 644534). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. The p.R180G variant (also known as c.538A>G), located in coding exon 4 of the POLD1 gene, results from an A to G substitution at nucleotide position 538. The arginine at codon 180 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
