This sequence change replaces arginine, which is basic and polar, with cysteine, which is neutral and slightly polar, at codon 322 of the POLD1 protein (p.Arg322Cys). To the best of our knowledge, the variant has not been reported in the published literature. The frequency of this variant in the general population, 0.0002 (6/29632 chromosomes in South Asian subpopulation, http://gnomad.broadinstitute.org), is higher than would generally be expected for pathogenic variants in this gene. The p.R322C variant (also known as c.964C>T), located in coding exon 7 of the POLD1 gene, results from a C to T substitution at nucleotide position 964. The arginine at codon 322 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
