Variant summary: POLD1 c.2669C>T (p.Ala890Val) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change. The variant allele was found at a frequency of 9.3e-05 in 247396 control chromosomes. The observed variant frequency is approximately 6.5 fold of the estimated maximal expected allele frequency for a pathogenic variant in POLD1 causing Colorectal Cancer phenotype (1.4e-05). To our knowledge, no occurrence of c.2669C>T in individuals affected with Colorectal Cancer or other POLD-related disorders and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 407969). Based on the evidence outlined above, the variant was classified as likely benign.
