The encoded protein, which contains an N-terminal pyrin domain, a NACHT domain, a NACHT-associated domain, and a C-terminus leucine-rich repeat region, functions as an attenuating factor of inflammation by suppressing inflammatory responses in activated monocytes. The NLRP12 c.1886C>A; p.Ala629Asp variant (rs146250162, ClinVar Variation ID 469090) is reported in the literature in an individual affected with common variable immunodeficiency (Borte 2014). This variant is found in the general population with an overall allele frequency of 0.011% (32/282808 alleles) in the Genome Aggregation Database (v2.1.1). Computational analyses predict that this variant is deleterious (REVEL: 0.905). Due to limited information, the clinical significance of this variant is uncertain at this time.
