This variant disrupts the p.Ala68 amino acid residue in RAB11B. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 29106825). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces alanine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 68 of the RAB11B protein (p.Ala68Ser). -
