This protein, like other neurexin proteins, contains epidermal growth factor repeats and laminin G domains. To our knowledge, this variant has not been reported in the literature or in a large population database (http://gnomad.broadinstitute.org), indicating this variant is rare. Los of function is not an established mechanism for CNTNAP5-related disease. However, a limited number of de novo deletions involving CNTNAP5 have been reported in individuals with autism and intellectual disability (Aleo et al 2020. PubMed ID: 32975021; Ludington et al 2020. PubMed ID: 32329157). Although we suspect that this variant may be pathogenic, at this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -
