This missense change has been observed in individual(s) with clinical features of hypomyelination with brain stem and spinal cord involvement and leg spasticity (PMID: 35571067). It has also been observed to segregate with disease in related individuals. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces alanine, which is neutral and non-polar, with glycine, which is neutral and non-polar, at codon 274 of the DARS protein (p.Ala274Gly).
