The observed missense c.703G>A (p.Glu235Lys) variant in ZEB2 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The p.Glu235Lys variant is absent in gnomAD Exomes. This variant has been submitted to the ClinVar database as Uncertain Significance. Multiple lines of computational evidence (Polyphen - Probably Damaging, SIFT - Damaging and MutationTaster - Disease causing) predict a damaging effect on protein structure and function for this variant. The reference amino acid of p.Glu235Lys in ZEB2 is predicted as conserved by GERP++ and PhyloP across 100 vertebrates. The amino acid Glu at position 235 is changed to a Lys changing protein sequence and it might alter its composition and physico-chemical properties. For these reasons, this variant has been classified as a Variant of Uncertain Significance (VUS). -
ClinVar contains an entry for this variant (Variation ID: 534634). This variant has not been reported in the literature in individuals affected with ZEB2-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces glutamic acid, which is acidic and polar, with lysine, which is basic and polar, at codon 235 of the ZEB2 protein (p.Glu235Lys). Advanced modeling performed at Invitae incorporating data from internal and/or published experimental studies (Invitae) indicates that this missense variant is not expected to disrupt ZEB2 function.
