Inborn genetic diseases Uncertain:1
The c.1136T>C (p.L379S) alteration is located in exon 14 (coding exon 13) of the ORC4 gene. This alteration results from a T to C substitution at nucleotide position 1136, causing the leucine (L) at amino acid position 379 to be replaced by a serine (S). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
not provided Uncertain:1
This sequence change replaces leucine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 379 of the ORC4 protein (p.Leu379Ser). This variant is present in population databases (rs780860667, gnomAD 0.009%). This variant has not been reported in the literature in individuals affected with ORC4-related conditions. ClinVar contains an entry for this variant (Variation ID: 211804). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
