Inborn genetic diseases Uncertain:1
The c.7076C>A (p.T2359N) alteration is located in exon 52 (coding exon 52) of the NBAS gene. This alteration results from a C to A substitution at nucleotide position 7076, causing the threonine (T) at amino acid position 2359 to be replaced by an asparagine (N). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
not provided Uncertain:1
This sequence change replaces threonine, which is neutral and polar, with asparagine, which is neutral and polar, at codon 2359 of the NBAS protein (p.Thr2359Asn). This variant is present in population databases (rs376484930, gnomAD 0.003%). This variant has not been reported in the literature in individuals affected with NBAS-related conditions. ClinVar contains an entry for this variant (Variation ID: 1378323). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
