Uncertain significance, criteria provided, single submitter | clinical testing | Ambry Genetics | Dec 08, 2024 | The p.R20C variant (also known as c.58C>T), located in coding exon 1 of the GEN1 gene, results from a C to T substitution at nucleotide position 58. The arginine at codon 20 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. - |

Uncertain significance, criteria provided, single submitter | clinical testing | Labcorp Genetics (formerly Invitae), Labcorp | Jul 16, 2021 | In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The cysteine amino acid residue is found in multiple mammalian species, suggesting that this missense change does not adversely affect protein function. These predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant has not been reported in the literature in individuals with GEN1-related conditions. This variant is present in population databases (rs778434447, ExAC 0.03%). This sequence change replaces arginine with cysteine at codon 20 of the GEN1 protein (p.Arg20Cys). The arginine residue is weakly conserved and there is a large physicochemical difference between arginine and cysteine. - |
