The Asp20471Glu variant in TTN has been identified by our laboratory in 1 Caucas ian infant with DCM (LMM unpublished data). This variant has not been identified in large European American and African American populations by the NHLBI Exome Sequencing Project (http://evs.gs.washington.edu/EVS), though it may be present in other populations. Computational analyses (biochemical amino acid properties, conservation, AlignGVGD, PolyPhen2, and SIFT) do not provide strong support for or against an impact to the protein. At this time, additional studies are neede d to fully assess the clinical significance of this variant. -

The Asp21398Glu variant in the TTN gene has not been reported as a disease-causing mutation or as a benign polymorphism, to our knowledge. Asp21398Glu results in a conservative amino acid substitution of one negatively charged amino acid for another. It is located in the A-band region of titin. The majority of mutations in the TTN gene are truncating mutations and these are located in the A-band region of titin (Herman D et al., 2012). The variant is found in TTN panel(s). -
