Variant summary: TTN c.44435A>T (p.Asp14812Val) results in a non-conservative amino acid change in the encoded protein sequence. Three of four in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 6.9e-05 in 247190 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in TTN causing Autosomal Recessive Titinopathy (6.9e-05 vs 0.00039), allowing no conclusion about variant significance. c.44435A>T has been reported in the literature in individuals with cardiomyopathy (example: Haskell_2017, Verhagen_2018, Roberts_2019). These report(s) do not provide unequivocal conclusions about association of the variant with Autosomal Recessive Titinopathy. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The p.D14812V variant (also known as c.44435A>T) is located in coding exon 222 of the TTNgene. This alteration results from an A to T substitution at nucleotide position 44435. The aspartic acid at codon 14812 is replaced by valine, an amino acid with highly dissimilar properties. Based on data from the NHLBI Exome Sequencing Project (ESP), the T-allele has an overall frequency of approximately 0.01% (1/11858), having been observed in 0.01% (1/8162) of European American alleles, and not observed in African American alleles studied.This variant was not reported in the Database of Single Nucleotide Polymorphisms (dbSNP) or the 1000 Genomes Project. Based on protein sequence alignment, this amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be possibly damaging by PolyPhen in silicoanalysis. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear.​ -
