The c.29848+6T>C variant in TTN has been identified by our laboratory in 1 infant with neonatal DCM and his affected father, both of whom also carried a likely pathogenic variant in MYH7. It was absent from large population studies. This variant is located in the 5' splice region. Computational tools do not suggest an impact to splicing. However, this information is not predictive enough to rule out pathogenicity. In summary, the clinical significance of the 29848+6T>C variant is uncertain. ACMG/AMP Criteria applied: PM2. -
