Likely pathogenic, criteria provided, single submitterclinical testingGeneDxAug 15, 2014c.528+5 G>C: IVS5+5 G>C in intron 5 of the COL3A1 gene (NM_000090.3) The c.528+5 G>C variant that is likely pathogenic was identified in the COL3A1 gene. Although the c.528+5 G>C variant has not been reported as a disease-causing mutation or as a benign polymorphism to our knowledge, this variant destroys the canonical splice donor site in intron 5 and is predicted to cause abnormal gene splicing. The variant is predicted to lead to either an abnormal message that is subject to nonsense-mediated mRNA decay, or to an abnormal protein product if the message is used for protein translation. Other splice site mutations in the COL3A1 gene have been reported in association with EDS type IV. Additionally, the c.528+5 G>C variant was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. Therefore, this variant is a strong candidate for a pathogenic mutation, however the possibility that it is a benign variant cannot be excluded. The variant is found in TAAD panel(s). -
