This sequence change replaces methionine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 202 of the STAT1 protein (p.Met202Val). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individuals with autosomal dominant chronic mucocutaneous candidiasis or cutaneous fusariosis (PMID: 21727188, 23245795, 24343863, 26604104). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 30087). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. Experimental studies have shown that this missense change affects STAT1 function (PMID: 21727188). For these reasons, this variant has been classified as Pathogenic. -
