The p.S3457C variant (also known as c.10370C>G), located in coding exon 26 of the APOB gene, results from a C to G substitution at nucleotide position 10370. The serine at codon 3457 is replaced by cysteine, an amino acid with dissimilar properties. This variant has been detected in a pediatric cohort with elevated total cholesterol (Klanar G et al. J Am Coll Cardiol, 2015 Sep;66:1250-1257). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
