This variant is present in population databases (rs763769862, gnomAD 0.001%). This missense change has been observed in individual(s) with dilated cardiomyopathy (PMID: 36396199). ClinVar contains an entry for this variant (Variation ID: 1311914). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. The p.E139Q variant (also known as c.415G>C), located in coding exon 1 of the DES gene, results from a G to C substitution at nucleotide position 415. The glutamic acid at codon 139 is replaced by glutamine, an amino acid with highly similar properties. This alteration has been reported in a dilated cardiomyopathy (DCM) cohort (Gigli M et al. J. Am. Coll. Cardiol., 2019 09;74:1480-1490). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
