PAX3 (HGNC:8617): (paired box 3) This gene is a member of the paired box (PAX) family of transcription factors. Members of the PAX family typically contain a paired box domain and a paired-type homeodomain. These genes play critical roles during fetal development. Mutations in paired box gene 3 are associated with Waardenburg syndrome, craniofacial-deafness-hand syndrome, and alveolar rhabdomyosarcoma. The translocation t(2;13)(q35;q14), which represents a fusion between PAX3 and the forkhead gene, is a frequent finding in alveolar rhabdomyosarcoma. The c.1174-3delT variant in PAX3 has not been previously reported in individuals with hearing loss, but has been identified in 1/10046 of South Asian chromosome s by the Exome Aggregation Consortium (ExAC, http://exac.broadinstitute.org; dbS NP rs749733628). Although this variant has been seen in the general population, its frequency is not high enough to rule out a pathogenic role. The c.1174-3delT variant is located in the 3' splice region. Computational tools do not suggest an impact to splicing. However, this information is not predictive enough to rul e out pathogenicity. In summary, the clinical significance of the c.1174-3delT v ariant is uncertain. -
