The c.1969C>T (p.R657C) alteration is located in exon 16 (coding exon 15) of the DIS3L2 gene. This alteration results from a C to T substitution at nucleotide position 1969, causing the arginine (R) at amino acid position 657 to be replaced by a cysteine (C). The p.R657C alteration is predicted to be tolerated by in silico analysis. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -

This sequence change replaces arginine, which is basic and polar, with cysteine, which is neutral and slightly polar, at codon 657 of the DIS3L2 protein (p.Arg657Cys). This variant is present in population databases (rs113330536, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with DIS3L2-related conditions. ClinVar contains an entry for this variant (Variation ID: 463083).
