Variant summary: CHRND c.236T>C (p.Ile79Thr) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 1.6e-05 in 251286 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.236T>C in individuals affected with CHRND-Related Disorders and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 1981765). Based on the evidence outlined above, the variant was classified as uncertain significance. -

This sequence change replaces isoleucine, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 79 of the CHRND protein (p.Ile79Thr). This variant is present in population databases (rs121909509, gnomAD 0.006%). This variant has not been reported in the literature in individuals affected with CHRND-related conditions.
