OTOF (HGNC:8515): (otoferlin) Mutations in this gene are a cause of neurosensory nonsyndromic recessive deafness, DFNB9. The short form of the encoded protein has 3 C2 domains, a single carboxy-terminal transmembrane domain found also in the C. elegans spermatogenesis factor FER-1 and human dysferlin, while the long form has 6 C2 domains. The homology suggests that this protein may be involved in vesicle membrane fusion. Several transcript variants encoding multiple isoforms have been found for this gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 64 amino acid(s) of the OTOF protein and extend the protein by 186 additional amino acid residues. -

The duplication causes a frameshift starting with codon Leucine 1934, changes this amino acid to a Proline residue and creates a premature Stop codon at position 251 of the new reading frame, denoted p.Leu1934ProfsX251. The c.5800dupC variant is not observed in large population cohorts (Lek et al., 2016). This likely pathogenic variant is predicted to result in protein truncation, as the last 64 amino acids are lost and replaced with 250 incorrect amino acids (Stenson et al., 2014). In summary, we interpret this variant as likely pathogenic. -
