This variant has not been published in the literature and is not present in population databases. This sequence change replaces aspartic acid with glutamic acid at codon 130 of the EPCAM protein (p.Asp130Glu). The aspartic acid residue is highly conserved and there is a small physicochemical difference between aspartic acid and glutamic acid. In summary, this is a novel missense change with uncertain impact on protein function. It has been classified as a Variant of Uncertain Significance. -

The p.D130E variant (also known as c.390C>G), located in coding exon 3 of the EPCAM gene, results from a C to G substitution at nucleotide position 390. The aspartic acid at codon 130 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
