The MSH2 p.Asp133= variant was identified in 4 of 5920 proband chromosomes (frequency: 0.0007) from individuals or families with HNPCC (Grabowski 2004, Mangold 2005, Samowitz 2001, Woods 2005). The variant was also identified in dbSNP (ID: rs61756462) as "With Uncertain significance allele", ClinVar (classified as benign by GeneDx; as likely benign by Invitae; as uncertain significance by two submitters), Mismatch Repair Genes Variant Database, and in Insight Hereditary Tumors Database (7x). The variant was not identified in COGR, Cosmic, MutDB, UMD-LSDB, or Zhejiang University databases. The variant was identified in control databases in 5 of 277186 chromosomes at a frequency of 0.00002 (Genome Aggregation Database Feb 27, 2017). The variant was observed in the European population in 5 of 126684 chromosomes (freq: 0.00004), while the variant was not observed in the African, Other, Latino, Ashkenazi Jewish, East Asian, Finnish, or South Asian populations. The p.Asp133= variant is not expected to have clinical significance because it does not result in a change of amino acid and is not located in a known consensus splice site. In addition, in silico or computational prediction software programs (SpliceSiteFinder, MaxEntScan, NNSPLICE, GeneSplicer) do not predict a difference in splicing.
