When cloned, it was discovered to be a human homolog of the E. coli mismatch repair gene mutS, consistent with the characteristic alterations in microsatellite sequences (RER+ phenotype) found in HNPCC. This sequence change replaces proline, which is neutral and non-polar, with serine, which is neutral and polar, at codon 349 of the MSH2 protein (p.Pro349Ser). -
