The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1347G>C has been reported in the literature in multiple individuals affected with various types of cancer (example: Whelan_2002, Tung_2015, Zhang_2015, Shirts_2016, Ollodart_2021) without strong evidence for causality. These report(s) do not provide unequivocal conclusions about association of the variant with Hereditary Nonpolyposis Colorectal Cancer. In a yeast functional assay, the variant had a significantly lower mutation rate than wild-type, though this result did not replicate in an individual fluctuation assay on this variant (Ollodart_2021).
