However, it has been observed in an individual with a myeloid malignancy (PMID: 31911633 (2020)), in individuals with breast cancer (PMIDs: 34445631 (2021) and 33471991 (2021), see also LOVD (http://databases.lovd.nl/shared)), as well as in reportedly healthy individuals (PMIDs: 36243179 (2022)) and 33471991 (2021), see also LOVD (http://databases.lovd.nl/shared)). A screening assay based on cell survival in response to 6-thioguanine treatment indicates that this variant has a neutral effect on DNA mismatch repair function (PMID: 33357406 (2021)), however, further studies are required to determine the global effect of this variant on MSH2 protein function. The frequency of this variant in the general population, 0.000008 (2/250906 chromosomes (Genome Aggregation Database, http://gnomad.broadinstitute.org)), is uninformative in the assessment of its pathogenicity. Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded conflicting predictions that this variant is deleterious or benign. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. Although reported in breast cancer cases and controls in settings of multigene panel testing (example, Dorling_2021), to our knowledge, c.1724A>G has not been reported in the literature in individuals affected with Hereditary Nonpolyposis Colorectal Cancer/Lynch syndrome. These report(s) do not provide unequivocal conclusions about association of the variant with Hereditary Nonpolyposis Colorectal Cancer. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
