The p.S586* pathogenic mutation (also known as c.1757C>G), located in coding exon 11 of the MSH2 gene, results from a C to G substitution at nucleotide position 1757. This changes the amino acid from a serine to a stop codon within coding exon 11. This alteration was detected in 1/132 unrelated individuals who met either Amsterdam I or Amsterdam II criteria (De Lellis L et al. PLoS ONE, 2013 Nov;8:e81194). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
