The variant was observed in the following populations: Latino in 1 of 33582 chromosomes (freq: 0.00003) and European Non-Finnish in 2 of 111674 chromosomes (freq: 0.00002), while it was not observed in the African, Ashkenazi Jewish, East Asian, European Finnish, Other, or South Asian populations. The p.Gln645 residue is located in the ATPase domain of the MSH2 protein (Kansikas, 2011) and is conserved in mammals; however, computational analyses (PolyPhen-2, SIFT, AlignGVGD, BLOSUM, MutationTaster) provide inconsistent predictions regarding the impact to the protein; this information is not very predictive of pathogenicity.
