This variant has not been reported in the literature in individuals with MSH6-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces leucine with arginine at codon 926 of the MSH6 protein (p.Leu926Arg). The leucine residue is weakly conserved and there is a moderate physicochemical difference between leucine and arginine. -

The p.L926R variant (also known as c.2777T>G), located in coding exon 4 of the MSH6 gene, results from a T to G substitution at nucleotide position 2777. The leucine at codon 926 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -
