The c.3887A>G (p.K1296R) alteration is located in exon 9 (coding exon 9) of the MSH6 gene. This alteration results from a A to G substitution at nucleotide position 3887, causing the lysine (K) at amino acid position 1296 to be replaced by an arginine (R). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
This missense variant replaces lysine with arginine at codon 1296 of the MSH6 protein. Computational prediction is inconclusive regarding the impact of this variant on protein structure and function (internally defined REVEL score threshold 0.5 < inconclusive < 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with MSH6-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD).
