The p.M1326I variant (also known as c.3978G>C), located in coding exon 9 of the MSH6 gene, results from a G to C substitution at nucleotide position 3978. The methionine at codon 1326 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
This missense variant replaces methionine with isoleucine at codon 1326 of the MSH6 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with MSH6-related disorders in the literature. This variant is denoted MSH6 c.3978G>C at the cDNA level, p.Met1326Ile (M1326I) at the protein level, and results in the change of a Methionine to an Isoleucine (ATG>ATC). This variant has not, to our knowledge, been published in the literature as pathogenic or benign. MSH6 Met1326Ile was not observed at a significant allele frequency in large population cohorts (Lek 2016). This variant is located in the binding site of MSH2 within the ATPase domain (Kariola 2002, Warren 2007, Kansikas 2011). In silico analysis, which includes protein predictors and evolutionary conservation, supports that this variant does not alter protein structure/function.
