The alanine at codon 186 is replaced by proline, an amino acid with highly similar properties. This alteration has been identified in multiple individuals diagnosed with pheochromocytoma (Ben Aim L et al. J Med Genet, 2019 Aug;56:513-520; Armaiz-Pena G et al. J Clin Endocrinol Metab, 2021 Jan;106:e350-e364). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
