The p.Ile155Phe variant in MYLK2 has been identified by our laboratory in 1 infa nt with mitral and tricuspid valve dysplasia. This variant has also been identif ied in 4/62662 of European chromosomes by the Exome Aggregation Consortium (ExAC , http://exac.broadinstitute.org; dbSNP rs140233643). Computational prediction t ools and conservation analysis suggest that the p.Ile155Phe variant may not impa ct the protein, though this information is not predictive enough to rule out pat hogenicity. In summary, the clinical significance of the p.Ile155Phe variant is uncertain. -
