Genes affected
ITCH (HGNC:13890): (itchy E3 ubiquitin protein ligase) This gene encodes a member of the Nedd4 family of HECT domain E3 ubiquitin ligases. HECT domain E3 ubiquitin ligases transfer ubiquitin from E2 ubiquitin-conjugating enzymes to protein substrates, thus targeting specific proteins for lysosomal degradation. The encoded protein plays a role in multiple cellular processes including erythroid and lymphoid cell differentiation and the regulation of immune responses. Mutations in this gene are a cause of syndromic multisystem autoimmune disease. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene. ACMG classification

Classification made for transcript

Verdict is Uncertain_significance. Variant got 3 ACMG points.

PM1
In a modified_residue N-acetylserine (size 0) in uniprot entity ITCH_HUMAN
PM2
Very rare variant in population databases, with high coverage;
PP2
Missense variant where missense usually causes diseases, ITCH
BP4
Computational evidence support a benign effect (MetaRNN=0.18504474).

Uncertain significance, criteria provided, single submitterclinical testingAmbry GeneticsDec 21, 2023The c.5C>G (p.S2C) alteration is located in exon 3 (coding exon 1) of the ITCH gene. This alteration results from a C to G substitution at nucleotide position 5, causing the serine (S) at amino acid position 2 to be replaced by a cysteine (C). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
