Variant summary
Our verdict is Uncertain significance. Variant got 2 ACMG points: 2P and 0B. PM2
The NM_030777.4(SLC2A10):c.848C>G(p.Ala283Gly) variant causes a missense change involving the alteration of a non-conserved nucleotide. The variant allele was found at a frequency of 0.00000137 in 1,461,846 control chromosomes in the GnomAD database, with no homozygous occurrence. 12/21 in silico tools predict a benign outcome for this variant. Variant has been reported in ClinVar as Uncertain significance (★).

Familial thoracic aortic aneurysm and aortic dissection Uncertain:1
The p.A283G variant (also known as c.848C>G), located in coding exon 2 of the SLC2A10 gene, results from a C to G substitution at nucleotide position 848. The alanine at codon 283 is replaced by glycine, an amino acid with similar properties. This variant co-occurred with a variant in the COL3A1 gene in an individual from a thoracic aortic aneurysm and dissection cohort; however, details were limited (Kathiravel U et al. Mol. Cell. Probes, 2013 Apr;27:103-8). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
