Variant summary
Our verdict is Uncertain significance. The variant received 3 ACMG points: 5P and 2B. PM2PM5PP2BP4_Moderate
The NM_172107.4(KCNQ2):c.2315C>G(p.Pro772Arg) variant causes a missense change involving the alteration of a non-conserved nucleotide. The variant allele was found at a frequency of 0.00000189 in 1,583,636 control chromosomes in the GnomAD database, with no homozygous occurrence. In-silico tool predicts a benign outcome for this variant. Variant has been reported in ClinVar as Uncertain significance (★). Another variant affecting the same amino acid position, but resulting in a different missense (i.e. P772L) has been classified as Likely pathogenic.

The c.2315C>G (p.P772R) alteration is located in exon 17 (coding exon 17) of the KCNQ2 gene. This alteration results from a C to G substitution at nucleotide position 2315, causing the proline (P) at amino acid position 772 to be replaced by an arginine (R). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
