This sequence change replaces aspartic acid with glutamic acid at codon 355 of the KCNQ2 protein (p.Asp355Glu). The aspartic acid residue is highly conserved and there is a small physicochemical difference between aspartic acid and glutamic acid. In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). This variant has been observed in individual(s) with neonatal seizures (PMID: 28926830, Invitae). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (ExAC no frequency). -
