Variant summary: C20orf54 c.1371C>G (p.Phe457Leu) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 0.00038 in 269176 control chromosomes (gnomAD). c.1371C>G has been reported in the literature in individuals affected with Brown-Vialetto Laere Syndrome without strong evidence of causality (Bansagi_2017, Green_2010, Johnson_2012, Manole_2017). These reports do not provide unequivocal conclusions about association of the variant with Brown-Vialetto Laere Syndrome 1. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 20206331, 22740598, 29053833, 28251916).
