This alteration results from a T to G substitution at nucleotide position 44, causing the leucine (L) at amino acid position 15 to be replaced by a tryptophan (W). This sequence change replaces leucine, which is neutral and non-polar, with tryptophan, which is neutral and slightly polar, at codon 15 of the IFNAR2 protein (p.Leu15Trp). This variant is present in population databases (rs755873190, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with IFNAR2-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0". The tryptophan amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function.
